Abdi Nageeye of the Netherlands was second in 2:09:58 with Belgium's Bashir Abdi finishing third in 2:10:00
Kenya's Lawrence Cherono came fourth in 2:10:02 while Amos Kipruto dropped out of the race at the 37 kilometers mark.
Kipchoge the world marathon record holder becomes the third athlete to defend an Olympic title in the marathon.
The Sunday race was characterized by sweltering heat with temperatures of 26 degrees Celsius and a 80 percent humidity.
